ABSTRACT

OBJECTIVES@#To study the efficacy and safety of domestic generic levetiracetam in replacement of brand-name levetiracetam in the treatment of children with epilepsy.@*METHODS@#A retrospective analysis was performed on the medical data of 154 children with epilepsy who received domestic generic levetiracetam in the inpatient or outpatient service of Guangdong Provincial People's Hospital from May 2019 to December 2020. Domestic generic levetiracetam and brand-name levetiracetam were compared in terms of efficacy and safety.@*RESULTS@#For these 154 children, the epilepsy control rate was 77.3% (119/154) at baseline. At 6 months after switching to domestic generic levetiracetam, the epilepsy control rate reached 83.8% (129/154), which showed a significant increase (P<0.05). There was no significant change in the frequency of seizures from baseline to 6 months after switching (P>0.05). The incidence of refractory epilepsy in children with no response after switching treatment was significantly higher than that in children with response (P<0.05). Before switching, only 1 child (0.6%) experienced somnolence, while after switching, 3 children (1.9%) experienced mild adverse drug reactions, including dizziness, somnolence, irritability, and bad temper.@*CONCLUSIONS@#Switching from brand-name to generic levetiracetam is safe and effective and holds promise for clinical application, but more prospective randomized controlled trials are required in future.

ABSTRACT

Objective: To explore the effect of a novel long-noncoding RNA (LncRNA) LOC90024 on cell viability, migration and invasion in A549 cells. Methods: The expression of LOC90024 in lung cancer cells A549 and normal lung cells BEAS-2B was initially screened by Real-time qPCR. The recombinant plasmid of LOC90024 was constructed. The effects of LOC90024 on cell proliferation, migration ability and cell invasion of A549 were detected by Real-time qPCR, MTT, Wound scratch assay and Transwell assay, respectively. Results: Recombinant plasmid with LOC90024 was successfully constructed. Compared with that in blank group, the expression of LOC90024 was significantly increased in overexpression group. MTT results showed that the absorbance value of overexpression LOC90024 group significantly increased in A549 cells as compared with that in blank group. Wound scratch assay showed that the scratch healing ability of overexpression LOC90024 group also significantly increased in A549 cells. Transwell experiment results showed that the number of cells that went through the membrane in overexpression LOC90024 group significantly increased and the A620 value was increased. Conclusion: Overexpression of lncRNA LOC90024 significantly increased cell viability, migration and invasion of A549 cells, indicating that it might function as an oncogene in lung cancer progression and has the prospect of becoming a new target for lung cancer treatment.

ABSTRACT

Objective To explore the effects of heart rate changes after holding breath and time for recovering stable heart rate on the quality of coronary CTA.Methods Totally 700 patients undergoing coronary CTA examination in some hospital were enrolled into the study,whose data on initial heart rate at rest condition,maximal heart rate during breath holding,stable heart rate after breath holding as well as the time consumed for recovering stable heart rate were collected and analyzed.Results A heart rate trendgram was drawn to find out the rules for heart rate changes and time for recovering stable heart rate,so that proper retrospective or prospective scanning scheme could be determined.Conclusion Mastering the rules in heart rate changes and time for recovering stable heart rate contributes to guiding coronary CTA.

ABSTRACT

<p><b>AIM</b>To observe the dynamic changes of heme oxygenase-1 (HO-1) mRNA and protein express in subfornical organ in rats with experimental allergic encephalomyelitis (EAE) to confirm that SFO is one of the sites for blood-bearing signaling molecules entering into brain.</p><p><b>METHODS</b>EAE was induced by CFA-GPSCH on Wistar rats, we observed the levels of HO-1 mRNA and its protein expression with immunohistochemistry and in situ hybridization technology on 1 d, 7 d, 14 d, and 21 d after EAE induction in SFO of rats. The relationship between HO-1 and symptoms of EAE was also investigated.</p><p><b>RESULTS</b>The expression levels of HO-1 mRNA and its protein expression were very low in the brains of the control group, whereas they were enhanced gradually with pathological course in the brain and onsets of symptoms, signs of EAE. On 1 d after induction of EAE, positive cells of HO-1 mRNA and its protein expression were observed at SFO, but the labeled cells were rarely seen in the other brain regions. On 7 d, the positive cells increased markedly. On 14 d the levels of HO-1 mRNA and its protein expression in the brains reached the peak, the positive cells of HO-1 were mainly located at the choroid plexuses and SFO, as well as the regions around "sleeve-like" lesion foci, all of which were coincident with the locations of lesions of EAE. The changes of incidence, symptom, reduction of the body weight, and pathology lesions of EAE in rat brains were the most significant. On 21 d, the levels of HO-1 mRNA and its protein expression reduced gradually, which was in parallel with remitted symptoms of EAE. When a specific inhibitor of HO-1, Snpp9, was applied, the symptoms and pathological lesions of EAE in brains were mitigated markedly.</p><p><b>CONCLUSION</b>SFO may be one of the earliest sites for blood-bearing signaling molecules entering into brain. The dynamic changes of HO-1 mRNA and its protein expression are in parallel with the changes of symptoms and pathological lesions of EAE in the brains. Application of some inhibitors of HO-1 may be one of potential therapeutic methods for prevention and treatment of EAE.</p>

@#Rehabilitation is very important to the therapeutic effect of total knee arthroplasty, and it's also the key measure for the knee function to reach the expected effect after the surgery. The most important contents of rehabilitation therapies are the training of muscle strength, range of joint motion, joint position sense and gait. Integrated rehabilitation therapies was used to improve the knee function as possible, such as all kinds of training to improve the knee muscle strength of flex and extend; early training of continued positive motion of knee joint to improve the knee range of motion; the training of knee joint position sense to improve the ability of motion control, posture adjusting and balance keeping; the gait training to adjust the unusual gait.

ABSTRACT

To define the action sites of adrenomedullin (ADM) in the rat brain, and to examine whether neuronal NO may participate in the actions of ADM, the present study was undertaken to examine the effects of i.c.v. administration of ADM on the induction of Fos protein and on nitric oxide-producing neurons in rat brain nuclei involved in cardiovascular regulation, using double immunohistochemical method for Fos and neuronal nitric oxide synthase (nNOS). Following i.c.v. administration of ADM (1 nmol/kg, 3 nmol/kg), Fos-like immunoreactivity neurons were markedly increased in several brain areas of the rat, including the nucleus of the solitary tract (NTS), the area postrema, the locus coeruleus, the parabrachial nucleus and the nucleus paragigantocelluaris laterialis (PGL) in the brainstem, the paraventricular nucleus (PVN), the supraoptic nucleus (SON) and the ventromedial hypothalamic nucleus in the hypothalamus, as well as the central amygdaloid nucleus and the lateral habenular nucleus in the forebrain. Following i.c.v. injection of ADM (1 nmol/kg, 3 nmol/kg), the number of double-labeled neurons for Fos and nNOS was increased in the PVN and SON. Small numbers of double-labeled neurons were also found in the NTS and PGL following i.c.v. injection of ADM (3 nmol/kg), while i.c.v. injection of ADM (1 nmol/kg) did not change the number of double-labeled neurons in the NTS and PGL. Pretreatment with calcitonin gene-related peptide receptor antagonist CGRP(8-37) (30 nmol/kg) significantly reduced the action of ADM (3 nmol/kg) in the brain. These results suggest that centrally administered ADM may increase the expression of c-fos in the forebrain, the hypothalamus and the brainstem and activate nitric oxide-producing neurons in the PVN, SON, NTS and PGL. These effects may be partly mediated by CGRP receptors.

@#ObjectiveTo study the effect of low-intensity millimeter wave(MMW)irradiation on the immune adhesion function of erythrocytes in mice with tumor.Methods60 healthy male mice were randomized into two groups:irradiation group(n=30) and control group(n=30).All mice were injected into abdominal cavity with A's abdominal juice cancer-cell(1×106/ml,0.2ml).MMW(36GHz,0.73-1.46mw/cm2)was used to irradiate the back of mice in irradiation group,once a day for 8 days.Control group received false irradiation using the same method.At 0d,3d,6d after irradiation, the erythrocytes were extracted from vein blood in the mice,and the RBC-C3b receptor rosettes rate(RCR),tumour-RBC rosette rate(TRR),RBC-SOD and RBC-LPO were determined.ResultsAt 3d after radiation,the RBC-C3b and TRR of irradiation group were higher than that of control group (P<0.01) and RBC-LPO was lower (P<0.05),while there was no significant difference on activity of RBC-SOD(P>0.05). there were no significant difference on all index between two groups at 0d and 6d(P>0.05).ConclusionsLow-intensity millimeter wave irradiation can improve the immune adhesion function of erythrocytes in mice with tumor.
